Can you make realistic people in Blender software?

I want to make a short movie with real people rather than "cartoonish" characters.

It is possible, you'll need time, skill and a powerful computer, but it is possible.

The difference between cartoony and realistic is usually just a case of (a) getting the proportions right (people tend to make the hands and head bigger then they should be, which looks cartoony) and (b) putting in enough detail (flat areas and smooth curves look less realistic).
